Notre étude menée sur une cohorte de jeunes hommes révèle que les athlètes d'endurance présentent un microbiote intestinal distinct, caractérisé par une augmentation de certaines espèces bactériennes et de métabolites spécifiques. Par ailleurs, une transplantation de microbiote fécal (FMT) de ces athlètes chez des souris améliore leur sensibilité à l’insuline et leurs réserves musculaires de glycogène, soulignant le rôle fonctionnel du microbiote dans la régulation du métabolisme énergétique. Ensuite, une nouvelle approche statistique est proposée afin d’explorer les signaux de faible amplitude (bactéries faiblement exprimées), souvent occultés par les espèces dominantes. Cette transformation permet d'améliorer la détection d’associations entre le microbiote et la physiologie de l'hôte, et optimise les performances des modèles prédictifs en apprentissage automatique. Notre approche améliore aussi l'interprétabilité des différentes méthodes d'analyses. This thesis aims to explore the interaction between the gut microbiota and host energy metabolism, and developing analysis methods adapted to metagenomic data. Our study including a cohort of young men reveals that endurance athletes have an atypical gut microbiota, characterized by an increase in specific bacterial species and specific metabolites. Furthermore, transplantation of fecal microbiota (FMT) from these athletes into mice improved insulin sensitivity and muscle glycogen stores, highlighting the functional role of microbiota in regulating energy metabolism. Next, a new data transformation is proposed to explore minor signals (i.e., weakly expressed bacteria), often masked by dominant species. This data transformation 1) enhances the detection of associations between the microbiota and host physiology, thereby improving the performance of machine learning-based predictive models, and 2) increases the interpretability of microbiome data analyses. The identification of these minor microbial signatures as relevant health indicators opens promising perspectives for the predictive modeling of metabolic disorders.
