Ce médicament engendre de nombreux effets indésirables dont le mécanisme reste méconnu. La littérature désigne le ratio AUC0-24h/CMI comme meilleur marqueur PK-PD d’efficacité pour le traitement des infections ostéoarticulaires. Cependant, aucune étude n’a recherché le lien entre l’exposition à la LVF, reflétée par son aire sous la courbe des concentrations sur 24h (AUC0-24h), et la survenue d’effets indésirables. Notre étude s’est intéressée à démontrer cette relation. Nos analyses ne montrent pas de différence significative des paramètres pharmacologiques chez les patients ayant développé un ou plusieurs effets indésirables par rapport à ceux n’en ayant pas développés. En revanche, nous montrons qu’il existe une corrélation entre la concentration résiduelle et l’exposition à la LVF. Ce paramètre pourrait devenir un biomarqueur intéressant pour la Lévofloxacine, permettant un suivi thérapeutique pharmacologique plus accessible. Bones and joins (BJ) infections are regularly treated with Levofloxacin (LVF). This drug is associated with a lot of adverse effects, the mechanism of which remains poorly understood. Literature identifies the AUC0-24h/CMI ratio as the best PK-PD marker of efficacy in the treatment of BJ infections. However, no study has investigated the link between exposure to LVF, reflected by its area under the 24-hour concentration curve (AUC0-24h), and the occurrence of adverse effects. Our study sought to demonstrate this relationship. Our analyses showed no significant difference in pharmacological parameters between patients who developed one or more adverse events and those who did not. However, we show that there is a correlation between residual concentration and exposure to LVF. This parameter could become an interesting biomarker for Levofloxacin, enabling more accessible therapeutic drug monitoring.
