     <dcterms:abstract xml:lang="fr">Le signal RMN de relaxation transversale de la feuille de colza permet de révéler la mise en place des changements structuraux et de distribution de l’eau dans le mésophylle au cours du développement foliaire. A la transition puits-sources, ces modifications sont susceptibles d’altérer la réponse adaptative au stress hydrique. L’objectif de la thèse était d’interpréter les variations du signal RMN et de comprendre l’effet de ces changements sur la régulation des flux d'eau en réponse à la sécheresse. En combinant des approches de relaxométrie RMN, diffusiométrie, IRM, physiologie, microscopie et multi-omiques, il a été possible de caractériser les structures cellulaires, les solutés impliqués dans l’ajustement osmotique, les relations hydriques à l’échelle des tissus ou de l’ensemble du limbe sur des plantes de colza en conditions hydrique optimales, stressées ou réhydratées. Les résultats obtenus ont montré le rôle de la plasticité structurelle et métabolique dans la réponse adaptative des feuilles. L’imagerie IRM a révélé l’hétérogénéité des structures et de l’état hydrique à l’échelle du limbe, tandis que les résultats de la mesure du coefficient d’autodiffusion vacuolaire soutiennent un accroissement de la teneur en eau dans les vacuoles des cellules du parenchyme palissadique à la transition puits-source et sa diminution en réponse au stress hydrique. Ces résultats pourront alimenter les modèles existants sur le fonctionnement hydraulique foliaire.</dcterms:abstract>
     <dcterms:abstract xml:lang="en">The transverse relaxation NMR signal from oilseed rape leaves reveals structural and water distribution changes in the mesophyll during leaf development. At the sink-to-source transition, these changes are likely to alter the adaptive response to water stress. The aim of this work was to interpret the variations in the NMR signal and to understand the impact of these changes on the regulation of water flux in response to drought. By combining NMR relaxometry, diffusiometry, MRI, physiology, microscopy and multi-omics approaches, it was possible to characterise the cellular structures, solutes involved in osmotic adjustment and water relations at the tissue or whole leaf scale in oilseed rape plants under optimal, stressed or rehydrated water conditions. The results demonstrated the role of structural and metabolic plasticity in the adaptive response of leaves. MRI imaging revealed the heterogeneity of structures and water status at the blade scale, while vacuolar self-diffusion coefficient measurements supported an increase in water content in the vacuoles of leaf parenchyma cells at the sink-to-source transition and its decrease in response to water stress. These results could feed models of leaf hydraulic functioning.</dcterms:abstract>
