     <dcterms:abstract xml:lang="fr">Ce manuscrit décrit une étude de chimie médicinale visant à développer un inhibiteur ciblant la protéine kinase GSK-3α, en vue de son évaluation sur des lignées cellulaires de cancers agressifs qui surexpriment cet isoforme. Ce travail s'inscrit ainsi dans la continuité des travaux initiés par notre équipe, visant à améliorer une nouvelle classe d'inhibiteurs basée sur un squelette oxazolo[5,4-f]quinoxaline. Nous avons ici synthétisé une bibliothèque de 88 composés parmi lesquels le composé le plus puissant (MH-124) s’avère 14 fois plus actif sur GSK-3α (IC₅₀ = 17 nM) que sur GSK-3β. De plus, nous avons développé une stratégie de synthèse permettant d’accéder à de nouveaux analogues comportant un système tricyclique phosphoré. En parallèle des efforts déployés pour identifier des inhibiteurs plus puissants et sélectifs de GSK-3α, nous avons entrepris de tester l’efficacité de MH-124 in vitro sur des cellules de cancers qui surexpriment GSK-3α, notamment en combinaison avec un traitement utilisé en clinique. Nous avons ainsi montré, sur deux lignées de glioblastome, l’intérêt de combiner notre inhibiteur avec un traitement au témozolomide, MH-124 resensibilisant les cellules de ce cancer au traitement.</dcterms:abstract>
     <dcterms:abstract xml:lang="en">This manuscript describes a medicinal chemistry study dedicated to the development of inhibitors targeting the GSK-3α protein, for evaluation against aggressive cancer cell lines overexpressing this isoform. In continuation of the work initiated in our team, the project is dedicated to the optimization of a new class of oxazolo[5,4-f]quinoxaline-based inhibitors. Here, we synthesized a library of 88 compounds, the most potent one (MH-124) being 14 times more active on GSK-3α (IC₅₀ = 17 nM) than on GSK-3β. Furthermore, we have developed a synthetic approach toward new analogues based on a phosphorus-containing tricyclic core. In parallel with these efforts to identify more potent and selective GSK-3α inhibitors, we also evaluated the in vitro efficacy of MH-124 against cancer cells overexpressing GSK-3α, both alone and in combination with a clinically used treatment. We have already demonstrated the benefit of MH-124/temozolomide combination, able to resensitize two glioblastoma cell lines to the treatment.</dcterms:abstract>
