     <dcterms:abstract xml:lang="fr">La différenciation terminale du lymphocyte B est un processus permettant de générer des plasmocytes sécréteurs d’anticorps, ainsi que des lymphocytes B mémoires. Ce processus est fortement régulé grâce à un ensemble de signaux présents dans le micro-environnement dont l'intégration détermine la progression du lymphocyte B vers le stade différencié. Au cours de ce projet, j'ai identifié que la signalisation IL-2 induit le programme de différenciation plasmocytaire par un mécanisme impliquant la voie de transduction ERK et le facteur de transcription BACH2. Ce travail apporte de nouvelles informations sur le rôle crucial de BACH2 dans le contrôle de la différenciation plasmocytaire humaine grâce, notamment, à des approches à l'échelle de la cellule permettant d'appréhender l'hétérogénéité des réponses immunes dans un modèle de différenciation in vitro. J'ai ensuite développé des études transcriptomiques sur cellules uniques pour une meilleure compréhension des mécanismes moléculaires spatiotemporels de la différenciation plasmocytaire normale d'une part, et des mécanismes moléculaires impliqués dans la pathologie de Waldenström résultant d'une différenciation plasmocytaire avortée d'autre part. Mes travaux valident la faisabilité technologique des approches expérimentales développées et ouvrent de nombreuses perspectives d'analyses visant à mieux caractériser la physiopathologie de la maladie de Waldenström.</dcterms:abstract>
     <dcterms:abstract xml:lang="en">Terminal B cell differentiation gives rise to antibody secreting plasma cells and memory B cells. It's a highly controlled process regulated by external inputs which guide B cell fate in the transition from naive B cells through to plasma cell. Here, we identify that IL-2 signaling triggers the plasma cell differentiation programme via the ERK transduction pathway instigating BACH2 repression. Results obtained using expression profiles of individual cells to bypass heterogeneous and asynchronous B cell responses confirm the crucial role of BACH2 regulating human B cell fate decision in a well-defined in vitro model of primary human naive B cell differentiation into plasma cell. My focus was then to study the temporal resolution of transcriptional dynamics following times series single-cell transcriptome analysis that should pinpoints key regulatory events that govern normal plasma cell differentiation in one hand and, on the other hand, the mechanisms that govern the differentiation arrest of plasma cell differentiation responsible for Waldenström disease, a rare incurable neoplasm. Altogether the data confirm the technological feasibility of the experimental strategies and should contribute to a better understanding of the complex mechanisms that control generation of plasma cell. This projec may ultimately help to achieve greater knowledge of the pathological process in Waldeström disease.</dcterms:abstract>
